干细胞因子在正常及梗死心肌中的表达Expression of stem cell factor in normal and infarct heart tissues

摘要(Abstract)：
目的 通过检测正常心肌组织及心肌梗死后干细胞因子 (SCF)mRNA的表达水平 ,为探讨其在心肌梗死中的作用奠定基础。方法 将成年SD大鼠随机分为正常组、结扎 6h组、结扎 1d组、结扎 3d组 (各时间点均设假结扎组以作对照 ) ,通过结扎左前降支建立心肌梗死模型 ,假结扎组只穿线不结扎。各组取心脏左室前壁同一部位 ,提取总RNA ,用RT PCR的方法 ,以GAPDH基因为内参 ,进行半定量检测正常心脏及梗死后SCFmRNA的表达水平。结果 可溶型SCF和膜型SCF在正常心肌有低水平表达。在心肌梗死后的三个不同时间点 ,结扎组心肌组织中可溶型SCF的表达分别为假结扎组的 1 6 6倍 (P <0 0 5 )、1 5 8倍和 1 30倍 (P <0 0 1) ;而膜型SCF的表达分别为假结扎组的1 80倍、1 5 3倍 (P <0 0 5 )和 1 5 7倍 (P <0 0 1)。结论 心肌梗死可以促使SCF的表达上调 ,提示其在心肌梗死后的病理过程中以及在诱导干细胞的归巢中可能发挥作用。
