摘要(Abstract):
目的观察188铼液体球囊血管内照射对细胞增殖、平滑肌细胞表型和转化生长因子的影响,探讨血管内照射预防再狭窄的可能机制。方法20只新西兰白兔随机分成照射组(n=10)和对照组(n=10),制作颈总动脉损伤模型。然后照射组以液体188铼液体球囊对动脉损伤处进行血管内照射,设定的照射吸收剂量为16Gy(参考点位于管腔表面下0.5mm);对照组以同样体积的生理盐水球囊,在动脉损伤处放置同样时间。30d后处死动物,取出颈总动脉作组织病理学、免疫组织化学及计算机图像分析。结果(1)照射组内膜中增殖细胞核抗原(proliferationcellularnuclearantigen,PCNA)、转化生长因子(transforminggrowthfactor,TGF)的免疫组化染色阳性指数明显小于对照组(0.028±0.005vs0.054±0.011,0.033±0.008vs0.063±0.014);(2)照射组和对照组内膜中平滑肌α肌动蛋白染色阳性指数差异无统计学意义(0.037±0.005vs0.032±0.003)。结论放射性同位素血管内照射可能通过抑制细胞分裂增殖、减少生长因子的分泌等机制预防血管介入治疗后再狭窄的发生;血管内照射可能不影响平滑肌细胞的表型。
