局部应用c-myc反义寡核苷酸对血管成形术后内膜增殖的影响Effects of local delivery c-myc antisense oligodexynucleotides on neointimal hyperplasia and vascular remodeling

摘要(Abstract)：
目的探讨局部转运cmyc反义寡核苷酸对兔腹主动脉粥样硬化狭窄球囊成形术后内膜增殖的影响。方法75只日本纯系雄性大耳白兔采用高胆固醇饮食加血管内皮剥脱的方法建立腹主动脉粥样硬化狭窄模型(狭窄>50%),并随机分为4组:反义治疗组(Ⅰ组)、正义治疗组(Ⅱ组)、生理盐水对照组(Ⅲ组)及单纯扩张组(Ⅳ)组。分别对各组狭窄段血管进行球囊扩张并进行血管内局部药物转运,分别于局部药物转运后24h及4周时取局部药物转运段血管,进行反转录PCR(RTPCR)半定量分析、cmyc蛋白免疫组化分析及组织形态学观察。结果局部药物转运后24h,Ⅰ组cmycmRNA水平明显低于Ⅱ、Ⅲ和Ⅳ组,局部药物转运后4周时,Ⅰ组新生内膜面积(NIA)、中膜面积(MA)及二者比值(NIA/MA)均明显小于Ⅱ、Ⅲ和Ⅳ组。结论经微孔球囊导管血管内局部应用cmyc反义寡核苷酸可抑制实验性兔腹主动脉粥样硬化狭窄球囊成形术后内膜增殖。
