阿托伐他汀对鼠颈总动脉血管损伤后CD40L和MMP-8表达影响Effcet of atorvastatin on the expression of MMP-8,CD40L and proliferation of intima in artery injured

摘要(Abstract):
目的观察在鼠颈总动脉血管损伤后血中CD40L、MMP-8水平和血管内膜增生情况以及他汀类药物干预后的影响。方法在鼠颈总动脉血管损伤模型上观察血管损伤及阿托伐他汀干预后的内膜增生情况,并应用ELISA方法检测血管损伤及阿托伐他汀干预后CD40L和MMP-8的血中浓度变化。结果与对照组比较,在鼠颈总动脉血管损伤后1周,血中CD40L和MMP-8浓度明显增高,分别为(0.97±0.11)ng/ml比(2.13±0.27)ng/ml,(9.12±1.39)ng/ml比(23.47±3.95)ng/ml,t值分别为2.13和23.47,P均<0.01。内膜明显增生(18.90±1.36)×103μm2比(69.13±4.38)×103μm2,F=16.145,P<0.01);2周时血中CD40L浓度降至(1.01±0.12)ng/ml,MMP-8浓度明显减弱为(12.16±1.45)ng/ml,而内膜增生更为明显(142.65±5.45)×103μm2。与手术损伤组比较,阿托伐他汀干预后血中CD40L和MMP-8浓度均明显下降,分别为(2.13±0.27)ng/ml比(1.37±0.19)ng/ml,(23.47±3.95)ng/ml比(14.48±2.08)ng/ml,t值分别为2.290和3.611,P均<0.05,内膜增生明显减轻(142.65±5.45)×103μm2比(81.76±4.35)×103μm2,F=12.737,P<0.05。结论鼠颈总动脉血管损伤后,内膜增生明显且CD40L和MMP-8血中浓度明显增高。血中CD40L和MMP-8浓度增高可能与平滑细胞迁移,新生内膜形成及血管重塑有关。阿托伐他汀可明显降低血中CD40L和MMP-8浓度,并减轻内膜增生。
